Résumé

Bien que l’utilisation des agents antiangiogéniques ait été considérée comme une stratégie prometteuse qui permet de bloquer la croissance tumorale et d’améliorer la biodisponibilité des médicaments jusqu’à la tumeur, l’utilisation en clinique de la plupart de ces agents s’avère limitée du fait du développement de la résistance à certains de ces antiangiogéniques et de leur importante toxicité. L’approche reste toutefois valable puisque cet outil thérapeutique a permis d’allonger la survie des patients sans progression pour ceux atteints de cancers coliques, mammaires, rénaux, pulmonaires et hépatiques. L’identification de marqueurs biologiques de réponse à cette famille de médicaments représente donc un domaine important d’investigation.

Abstract

Although the use of anti-angiogenic agents has been considered a promising strategy to block tumor growth and improve the bioavailability of drugs into the tumor, the use of most of them in clinical trials is limited. The development of resistance to some anti-angiogenic agents and their high toxicity are currently under investigations. However, the approach is still valid since this therapeutic tool has lengthened survival of patients with colon, breast, kidney, lungs and liver cancers. The identification of biomarkers in response to this family of drugs is an important area of investigation.
